A high-performance thin layer chromatography (HPTLC) method for the simultaneous quantitative determination of Lupeol and β- sitosterol in methanolic fraction of Achyranthes aspera leaves was developed. The separation of phytoconstituent was achieved using a mobile phase of toluene: ethyl acetate: formic acid (8:2:0.1, v/v/v). The densitometric determination was carried out at 550 and 525 nm in reflection/absorption mode for Lupeol and β-sitosterol. The calibration curves constructed in the range of 100-600 ng per spot for Lupeol and β- sitosterol were linear. HPTLC analysis of methanolic fraction of Achyranthes aspera leaves displayed the presence of Lupeol (0.09±0.04) and β- sitosterol (0.06±0.11). The developed method is simple, precise, specific, accurate, less time consuming and cost effective. The reproducibility and selectivity of method was assessed through statistical analysis of data. Thus, present method is established for simultaneous quantification of these compounds in Achyranthes aspera leaves which may be utilized for the fingerprinting and standardization of Achyranthes aspera .
